Lace plants explain programmed cell death

Programmed cell death (PCD) is a highly regulated process that occurs in all animals and plants as part of normal development and in response to the environment. New research published in BioMed Central's open access journal BMC Plant Biology is the first to document the physiological events in the lace plant (Aponogeton madagascariensis) which occur via PCD to produce the characteristic holes in its leaves.

The aquatic lace plant, endemic to Madagascar, uses PCD to generate holes in its leaves. Researchers from Dalhousie University, Nova Scotia, used long-term live cell imaging, time-lapse micro-photography and detailed staining to see what actually happens inside individual cells of the lace plant leaves during this process.

PCD always begins in the centre of areoles (areas surrounded by veins) and works its way out, finally stopping four or five cells away from the vein. Within these areoles the stage of PCD can be visually determined by pigmentation. Non-PCD cells contain anthocyanin which is pink. As PCD progresses (early-PCD) the anthocyanin disappears but green chlorophyll remains. By late-PCD, both of these pigments have been lost.

Arunika Gunawardena, who led this research, described what actually happens in more detail, "After the loss of anthocyanin, we saw a reduction in the number and size of chloroplasts concurrent with changes in the actin cytoskeleton, which normally supports the cell from the inside. Actin cables became thicker, but their arrangement became more haphazard (until late-PCD when they began to disappear)."

As PCD progressed chloroplasts and mitochondria (the energy centres of the cell) began to aggregate together and were brought into the acidic vacuole to be degraded - a process that was confirmed using 3D image analysis. Additionally, the researchers found evidence of autophagy (where the cells literally begin to 'eat' themselves from the inside) and discovered organelle-like remnants inside vesicles.

Dr Gunawardena continued, "For each cell we observed, eventually the tonoplast ruptured, and the plasma membrane surrounding the cell collapsed. It only takes 48 hours from the first noticeable loss in chlorophyll until the plasma membrane shrinks, and by 24 hours after this event the cell wall disappears."

This is the first research to use such a variety of visual techniques to look at a process that affects almost every cell type at some stage of an organism's life. These pictures and videos provide a fascinating insight into how the lace plant has high-jacked PCD to produce its quirky, yet beautiful, leaves.

Violence rages across Syria as UN mission clock ticks

Heavy clashes raged between Syrian troops and rebels on Saturday, with at least 130 people killed nationwide, as the clock ticked down on a 30-day "final" extension of a troubled UN observer mission.

At least 64 civilians were among the dead as clashes rocked both of the country's largest cities, the Syrian Observatory for Human Rights said.

Activists reported fierce fighting in the country's second city Aleppo, with regime forces shelling several neighbourhoods, as at least 12 civilians were reported dead in the capital, seven of them killed by snipers.

The violence came a day after the Security Council voted unanimously to grant a "final" 30-day extension to the UN Supervision Mission for Syria charged with overseeing a tattered peace plan.

UN chief Ban Ki-moon said he was "deeply distressed by the rising death toll," warning that the limited extension of the UN mission showed "the onus is above all on the parties, with the Syrian government in the first place, who must stop the killings."

The Syrian Observatory reported fierce fighting in several districts of Aleppo, including in the central Salaheddin district, although it had few immediate details on casualties.

"Violent clashes are taking place between Syrian regime forces and rebel fighters in the Sakhur neighbourhood and the Haydariya area," the Britain-based watchdog said, adding troops were "using heavy machineguns and artillery."

In Damascus, residents said the city was largely calm after days of heavy clashes, although fighting was reported in Al-Hajar Al-Aswad and Tadamon districts.

Among the 12 dead were a couple and their son, gunned down in Bab Tuma, a previously quiet Christian neighbourhood of the Old City.

In the flashpoint central city of Homs, government forces put down a nearly 24-hour mutiny at the main prison with two inmates found dead, the Observatory said.

It had no immediate word on the circumstances of the prisoners' deaths but earlier an activist told AFP he feared "a massacre" at the facility, which had been surrounded by tanks after several guards defected and prisoners took over one of its two wings.

The pace of violence in the country has ramped up in recent days, particularly after a Wednesday bombing -- claimed by the rebel Free Syrian Army -- which killed four senior regime members, including national security chief General Hisham Ikhtiyar, who died of his wounds on Friday.

The others killed in the blast, Defence Minister General Daoud Rajha, Assad's brother-in-law Assef Shawkat and General Hassan Turkmani, head of the regime's crisis cell on the uprising, were given a state funeral in Damascus on Friday.

The nationwide death toll the day after the attack stood at 302, the highest of the 16-month uprising, the Observatory said.

The Syrian army was battling on Saturday to retain control of the country's borders.

Iraqi officials said rebel forces had held onto the Albu Kamal crossing on the Euphrates river after capturing it on Thursday, and fighters had retaken the Rabiyah crossing on the main highway between Aleppo and Iraq's main northern city Mosul.

A third crossing between the two countries was still controlled by regime forces.

Syrian troops also lost control of the Bab al-Hawa crossing on the border with Turkey. An AFP photographer reported that it was in the hands of jihadists.

Turkish truck drivers accused the rebel Free Syrian Army of turning a blind eye while their vehicles were looted, The governor of the neighbouring Turkish province of Hatay, Celalettin Lekesiz, said "independent groups" and not the mainstream rebels were to blame.

In Jordan, a security official told AFP that Free Syrian Army fighters had tried to overrun the Nassib border crossing, but were repulsed by government troops.

Observatory director Rami Abdel Rahman told AFP: "The authorities now only really control some 50 to 60 percent of the country."

UN Security Council permanent members Russia and China on Friday both voted in favour of a resolution extending the mandate of UNSMIS for a "final" 30 days, after blocking an earlier text that could have imposed sanctions.

The resolution says the council could consider a further extension if violence reduces sufficiently to allow the mission "to implement its mandate."

US ambassador Susan Rice said the resolution would allow the observers "to withdraw safely and orderly" but that it was "unlikely" that the violence would ease enough to allow a continued UN presence.

But Russian ambassador Vitaly Churkin insisted: "This is not a resolution about withdrawal, it is a resolution about continuation of the activity of the mission."

The UN secretary general said he had sent peacekeeping chief Herve Ladsous to join chief military adviser General Babacar Gaye in Damascus to "lead UNSMIS in this critical phase."

Between a shaky global economy and diminishing demand for personal computers, hardware and software, investors in the tech industry are preparing for a wave of diminishing returns on account of weak earnings reports among companies in this field.

Facebook has announced that its revenue growth slowed down in the second quarter of 2012 and Yahoo?s performance has been affected by the firing of CEO Scott Thompson for lying on his resume. Component manufacturers such as Advanced Micro Devices, Seagate Technologies and Applied Materials each have given warnings that their sales figures would be lower than projected, with AMD in particular citing economic troubles in Europe and China and the reduced demand for personal computers.

However, despite the projected downturn, analysts say that the figures are not as bad as those reported in 2008. In addition, multi-national enterprise software manufacturer SAP reported that its second-quarter earnings jumped up eighteen per cent from 2011. Because SAP, headquartered in Germany, sells to companies worldwide, many investors and analysts were relieved by the company?s reports.

Hidden portals in Earth's magnetic field

ScienceDaily (July 3, 2012) ? A favorite theme of science fiction is "the portal" -- an extraordinary opening in space or time that connects travelers to distant realms. A good portal is a shortcut, a guide, a door into the unknown. If only they actually existed....

It turns out that they do, sort of, and a NASA-funded researcher at the University of Iowa has figured out how to find them.

"We call them X-points or electron diffusion regions," explains plasma physicist Jack Scudder of the University of Iowa. "They're places where the magnetic field of Earth connects to the magnetic field of the Sun, creating an uninterrupted path leading from our own planet to the sun's atmosphere 93 million miles away."

Observations by NASA's THEMIS spacecraft and Europe's Cluster probes suggest that these magnetic portals open and close dozens of times each day. They're typically located a few tens of thousands of kilometers from Earth where the geomagnetic field meets the onrushing solar wind. Most portals are small and short-lived; others are yawning, vast, and sustained. Tons of energetic particles can flow through the openings, heating Earth's upper atmosphere, sparking geomagnetic storms, and igniting bright polar auroras.

NASA is planning a mission called "MMS," short for Magnetospheric Multiscale Mission, due to launch in 2014, to study the phenomenon. Bristling with energetic particle detectors and magnetic sensors, the four spacecraft of MMS will spread out in Earth's magnetosphere and surround the portals to observe how they work.

Just one problem: Finding them. Magnetic portals are invisible, unstable, and elusive. They open and close without warning "and there are no signposts to guide us in," notes Scudder.

Actually, there are signposts, and Scudder has found them.

Portals form via the process of magnetic reconnection. Mingling lines of magnetic force from the sun and Earth criss-cross and join to create the openings. "X-points" are where the criss-cross takes place. The sudden joining of magnetic fields can propel jets of charged particles from the X-point, creating an "electron diffusion region."

To learn how to pinpoint these events, Scudder looked at data from a space probe that orbited Earth more than 10 years ago.

"In the late 1990s, NASA's Polar spacecraft spent years in Earth's magnetosphere," explains Scudder, "and it encountered many X-points during its mission."

Because Polar carried sensors similar to those of MMS, Scudder decided to see how an X-point looked to Polar. "Using Polar data, we have found five simple combinations of magnetic field and energetic particle measurements that tell us when we've come across an X-point or an electron diffusion region. A single spacecraft, properly instrumented, can make these measurements."

This means that single member of the MMS constellation using the diagnostics can find a portal and alert other members of the constellation. Mission planners long thought that MMS might have to spend a year or so learning to find portals before it could study them. Scudder's work short cuts the process, allowing MMS to get to work without delay.

It's a shortcut worthy of the best portals of fiction, only this time the portals are real. And with the new "signposts" we know how to find them.

Scientists at the University of East Anglia (UEA) have discovered a 'constant cloud' of potent inflammatory molecules surrounding the cells responsible for diseases such as thickening of the arteries and rheumatoid arthritis.

Published online today by The Journal of Cell Science, the findings could eventually lead to new treatments for chronic inflammatory diseases. Cardiovascular disease arising from atherosclerosis (thickening of the arteries) kills around 17 million people worldwide each year, including 120,000 people in England and Wales, while rheumatoid arthritis affects around 400,000 people in the UK.

The UEA team studied a type of white blood cell called monocytes. Monocytes play an important role in the human immune system and help protect our bodies against infection. But they can also invade tissue, triggering the early stages of common inflammatory diseases.

The researchers detected for the first time that monocytes were surrounded by a constant cloud. This cloud was found to be made up of potent inflammatory molecules called adenosine triphosphate, or ATP. Further study showed that the ATP molecules were being propelled through the cell wall by the actions of lysosomes. Lysosomes are sub-cellular compartments within blood cells which had previously been thought to only break down cell waste.

"These unexpected findings shed light on the very early stages in the development of inflammatory diseases such as atherosclerosis and rheumatoid arthritis," said lead author Dr Samuel Fountain of UEA's School of Biological Sciences.

"We found that lysosomes are actually highly dynamic and play a key role in the way inflammatory cells function. This is an exciting development that we hope will lead to the discovery of new targets for inflammatory drugs in around five years and potential new treatments beyond that."

Dr Fountain said further study was now needed to investigate how to control the release of ATP by lysosomes in monocytes and other white blood cells, and to understand how inflammation may be affected in patients with inherited diseases involving lysosomes.

Dr Fountain is a Biotechnology and Biological Sciences Research Council (BBSRC) David Phillips Fellow and recently received 0.9m from the BBSRC to study how cells use ATP as a signalling molecule.

Diablotek Abyss


There's been a heartening renaissance with budget PC cases in recent years. As models like the Thermaltake Dokker and the NZXT H2 have shown, it's possible for companies to include big features and deliver good build experiences, but still maintain a sub-$100 price. Unfortunately, these advancements are not universal. With its Abyss ($64.99 list), Diablotek demonstrates that it's possible to get the broadest strokes correct but drop the ball on practically everything else. The result is a case that fulfills its basic task, but does so with little finesse.

The Abyss' best quality is, in fact, its outward flair. The 17.2-by-8.3-by-20-inch (HWD) case is fashioned mostly of white steel, with three significant exceptions: the white plastic front panel, the black mesh ornamentation on that panel (specifically, the covers over the drive bays and the 120mm intake fan); and the windowed side panel. It's not necessarily an adventurous look, but plenty of clever angles (the fan grille resembles a star, for example), unexpected lines, and vent-shaped depressions help the Abyss project a healthy sci-fi vibe that makes it appear more expensive than it actually is. Two grommeted holes in the rear panel (where there's also a single 120mm exhaust fan) for routing liquid cooling piping also suggested latent gaming PC potential.

Open up the Abyss and you'll see what, at first glance, looks like a typically well-appointed interior (which, for the record, is primarily white as well). Two filtered vents in the floor (including one in the power supply bay) aid with passive cooling, and a large hole cut into motherboard tray's upper-left section facilitates adding an aftermarket CPU cooler, but there aren't that many additional holes for routing cables. Whereas Thermaltake and NZXT have lately begun giving you plenty of these, only four are accessible after you install an ATX motherboard on the Abyss: One, located near the bottom, is decently wide, but three others on the side are narrow and may make it difficult to send cables and wires where you want them. (If you're installing a microATX or Mini ITX motherboard, you will have two more options, but they're even smaller.) And, aside from the channel those holes delineate, you'll find almost no room below the motherboard for stashing cables.

You do have a generous amount of space (slightly more than 1.5 inches) between the motherboard's interior edge and the start of the drive well, though some of this will be compromised when you add 3.5-inch drives (the intake fan keeps them from sliding all the way in). But there's only one-eighth inch between the bottom of the motherboard and the top of the power supply?cramped quarters for connecting cables to headers or hooking up front-panel wires. Space is a problem from another standpoint, too: Although there's technically room to install an oversized video card like the AMD Radeon HD 6990, the case's interior design makes it impossible to do so in the most likely location of the first accessible PCI Express (PCIe) x16 slot, so even the attempt is pointless. It looks like it should work, but it doesn't quite.

That's true of numerous other elements, too. Eight of the drive bays (the four 5.25-inch external bays and the four 3.5-inch internal bays) use tool-free retention brackets to hold the drives in place, but having one on each side inconveniently forces you to open both side panels rather than just the main one to secure the components. The remaining two bays (one external 3.5-inch and the bottom-most internal 3.5-inch) require screws to keep the drives in place. And if you want to install a 2.5-inch drive but you don't have a 3.5-inch adapter, forget it: There's no place to even just screw in smaller drives.

The seven expansion slots are also not tool-free. The spacers are attached and pop out easily, but installing a card requires screwing it in?and, if you change your configuration later and want to replace one or more spacers, you can't even do that. This obviously saves money, but it's a poor solution; the inexpensive ($59.99 list) MSI Raptor's external bracket, which haphazardly clamped down its loose spacers, wasn't an ideal solution, but it was better (though it also required a screwdriver).

Wackier still are two other characteristics of the drive well and one of the front-panel ports. The fourth 5.25-inch bay is blocked by a mount for installing a second intake fan; there's simply no way to install an optical drive in it (though it might be able to house a smaller device of some kind), leaving one wondering why there's a bay there at all. Even stranger is the black plastic panel at the rear of the 3.5-inch bay section. Printed with the words "Remove for long card," it's precariously positioned between the second and third bays, but provided no discernible function as it doesn't block anything near even the longest cards. If you remove it, not only do you gain no appreciable component space, but you also subject yourself to the mother of all migraines trying to get back in its original position. (It took us?no exaggeration?ten minutes to properly align everything again.) And regarding the front-panel ports, it's nice to have two USB 3.0 ports accompanying the headphone and microphone jacks, but why is there a slot (even bracketed over) to accommodate an SD card reader when no reader is included?

Things like this don't identify the Diablotek Abyss as a serious case for serious builders. As it is, it will suffice provided your needs aren't extensive or your expectations high. But more satisfying cases are out there, for not much more money, in the aforementioned Thermaltake Dokker and NZXT H2. And even the MSI Raptor, which will save you a few dollars more, is less frustrating to build in. Diablotek's case may be a looker, but, as always, the devil is in the details, and that's where the Abyss derails.

Mexico's America Movil completes share bid for Dutch KPN


Mexican telecoms giant America Movil announced Thursday it had completed an unsolicited bid for a 27.7 percent share in Dutch operator KPN.

America Movil will now "hold 393,283,000 shares, representing approximately 27.7 percent of all issued and outstanding shares," it said in a statement.

Controlled by billionaire Carlos Slim, America Movil launched its bid on May 30 for just over a quarter of the Dutch group, offering eight euros ($10.07) a share or 2.6 billion euros ($3.2 billion) for the stake.

They gradually edged closer to the 27.7 percent goal, which was opposed by KPN who said it substantially undervalued the company.

"With this investment America Movil has achieved its objective of securing a meaningful minority stake in KPN, a company with an attractive position in important European markets," the statement said.

America Movil added: "Pursuant to this announcement... payment of the offer price for each tendered share accepted under the offer is made against delivery of such shares... which shall be July 3, 2012."

America Movil controls the mobile telephone market in Latin America with some 225 million clients in 18 countries at the end of 2010. In 2011, it posted the equivalent of 39 billion euros in revenues.

Dutch group KPN employs 31,000 people and recorded sales of 13.16 billion euros last year.

How sweet it is: Tomato researchers discover link between ripening, color and taste

ScienceDaily (June 28, 2012) ? For many grocery shoppers, those perfect, red tomatoes from the store just can't match the flavor from the home garden. Now, researchers at Boyce Thompson Institute for Plant Research at Cornell University, USDA and the University of California at Davis have decoded a gene that contributes to the level of sugar, carbohydrates and carotenoids in tomatoes.

Cuong Nguyen, a Cornell graduate student in plant breeding working at the Boyce Thompson Institute (BTI), along with colleagues at BTI, USDA, UC Davis, Universidad Politec?nica de Valencia (Spain), Universidad de M?laga (Spain) and University of Suleyman Demiral (Turkey) revealed the gene that underlies the uniform ripening mutation.

This gene also influences how tomato fruits ripen and is used by commercial breeders to create tomatoes that develop into perfectly red, store-ready fruit. "Practically, it is a very important trait," says James Giovannoni, a plant molecular biologist with BTI and the U.S. Department of Agriculture, Agricultural Research Service, who is a senior author on the paper. "It's a gene that whether you realize it or not, most of your tomatoes have." However, this same trait reduces sugars and nutrients in the fruit.

Naturally, tomatoes have uneven ripening, showing darker green patches when unripe and variable redness when ripe -- traits that still show up in garden-variety and heirloom breeds. However, in the late 1920s, commercial breeders stumbled across a natural mutation that caused tomatoes to ripen uniformly -- from an even shade of light green to an even shade of red. This mutation, known by plant biologists as 'uniform ripening', has become indispensable to the $2 billion a year US commercial tomato market, showing up in almost all tomatoes produced for grocery stores. The uniform redness makes it ideal for the grocery sector, which has to appeal to customer expectations of evenly colored, red fruit.

Nguyen conducted positional cloning and, with access to solgenomics.net, an online, public database hosted at BTI, he determined that the uniform ripening gene was located at a specific location on chromosome 10. With this location now known, the team could decipher the gene coding for the protein that controls photosynthesis levels in tomato fruit. While leaves are the primary photosynthesis factories in a plant, developing tomato fruit can contribute up to 20 percent of their own photosynthesis, yielding high sugar and nutrient levels in fully ripe fruit. The uniform ripening mutation, which commercial breeders select for, eliminates this protein in the fruit, therefore reducing sugar levels. "This is an unintended consequence," says Giovannoni, explaining why commercial growers continued to select for the trait. "Producers currently don't get a penny more for [flavor] quality."

This discovery has practical applications. Commercial producers -- who wish to produce uniform red fruit over multi-colored, flavorful ones -- can now do an early test on seedling DNA for the uniform ripening mutation, rather than waiting to observe the mature fruit. Conversely, those who don't care about appearances can make sure of the opposite -- that their plants are mutation free and thus may have better-tasting fruit.

Ann Powell, a research biochemist who led the UC Davis team's efforts on the research, says that the study "is a rare chance to translate scientific findings to the real world?it provides a strategy to re-capture quality characteristics that had been unknowingly bred out of modern cultivated tomatoes."

Funding for the Giovannoni lab at BTI is supported by the USDA and the National Science Foundation.
